The Blues have won just once in three previous 'Judgement Day' matches - against Scarlets in 2014 - but, in an eighth-versus-ninth clash, Wilson's men will be battling with Ospreys for a potential top six finish and to be the second-placed Welsh team behind Wayne Pivac's side.
Wales winger James, whose form for Blues since rejoining at the start of the season earned him an international recall from Warren Gatland during the Six Nations, hopes the packed national stadium can create an atmosphere to rival his days in the red of Wales.
"This is my first Judgement Day. It will be a bit different, a different atmosphere, only with Wales have I played in front of 72,000 and if we can get that attendance it will be awesome," the 29-year-old winger said.
"That's why you play rugby, for atmospheres like that, all the fans will be out in force and there will be a big buzz."
The Blues will be without Lloyd Williams and Rhys Patchell, but James, who has crossed the whitewash in the Guinness PRO12 on six occasions this season, is confident they can continue their winning league form.
"It's a good run we are on but we haven't beat the Ospreys for six years so that is another thing we want to do this year - beat them," he said.
"Hopefully we can do this on a regular basis. If we can play well the fans will turn up week in, week out. If we carry on as we are then they will come out all the time."
