Faced with a trip to Ireland to face Connacht in their final game, Glasgow will be hoping they can overhaul either of rivals Ulster or Leinster to secure a home semi-final.
Glasgow have twice reached the semi-final stage, in its first year of the new play-off format in 2009-10 and then last season, but lost both times away to the Ospreys and Leinster respectively.
No away semi-finalist has progressed to the final in the past three years but Cusiter, who won't feature until next season as he recovers from a long-term shoulder injury, insists the Warriors boast a squad capable of beating anyone home or away.
"The squad has changed a lot and I absolutely believe that they could win a semi-final home or away now," Cusiter told The Scotsman. "They have shown that they can beat any team in the league.
"It comes down to two big games really and, in the past, we have got over that line and made the top four but haven't been able to beat Leinster or Ospreys in the semi-finals, so that's what has to happen this year.
"But I do think that we're better equipped for it because this is a stronger squad than we've ever had and they guys are playing with a lot of confidence, and playing good rugby.
"History tells you you're better to have a home semi in three years of play-offs, but, in the situation we're in now, there's a big likelihood that we won't have a home semi-final, so we're going to be going to Ulsteror Leinster trying to beat them, but I don't think that's an impossible task."
