Tries from Keith Earls and Peter O'Mahony ensured the two-times European champions wrapped up top spot in Pool 6 with one game to go.
Ian Keatley also kicked ten points as the current RaboDirect PRO12 leaders eased to victory - their dominant pack doing most of the damage.
Ulster are also guaranteed a spot in the quarter-finals despite missing out on a winning bonus point against Montpellier.
Tries from Robbie Diack, Ruan Pienaar and John Afoa helped them on the way to the 27-16 victory on Friday night to leave them in pole position in Pool 5.
And with Toulon beating Cardiff Blues 43-20 on Saturday, it means Ulster will qualify either as pool winners or one of the best two runners up.
The Blues themselves were outdone by the power of the French outfit's pack, conceding three penalty tries, while Jonny Wilkinson tormented them with 23 points from the kicking tee.
Filo Paulo and Sam Hobbs crashed over for the visitors and lying second in the group, they will now target an Amlin Challenge Cup place when they host Exeter Chiefs next week.
Three-times European Cup winners Leinster need just one point from next weeks game with Ospreys to qualify for the quarter finals after defeating Castres 29-22.
Jimmy Gopperth justified his selection over Ian Madigan at fly-half by scoring two tries, as his club came from 14-0 behind, while Jordi Murphy scored their third try.
Meanwhile Kristian Phillips' spectacular try helped Scarlets record their first win since November as they defeated Racing Metro 19-13 in Pool 4.
It means they also remain in with a shout of qualifying for the quarter-finals of the Amlin Challenge Cup, alongside Scottish pair Edinburgh and Glasgow Warriors who beat Perpignan 27-16 and Exeter Chiefs 15-10 respectively.
Connacht similarly kept alive their chances of progressing through in the Challenge Cup after beating fellow RaboDirect PRO12 outfit Zebre 20-3, as did Ospreys, despite losing 17-29 to Northampton Saints.
Elsewhere in the European Cup, Benetton Treviso lost out at the hands of Leicester Tigers 34-19 while in the pool stages of the Challenge Cup, Newport Gwent Dragons were defeated 30-13 by Bath.
TRY OF THE WEEKEND
A first win since November was a welcome plus for Scarlets from their game against Racing Metro but it was the sight of Kristian Phillips reducing the Racing defence to statues that was the highlight.
Gathering the ball high inside the Racing half, the right wing danced his way through their defence at devastating pace with just 19 minutes to go.
With Rhys Priestland converting, it put his side in the driving seat as they secured a gutsy win.
BREAK OF THE WEEKEND
Much of Edinburgh's win over Perpignan was built upon their solid defensive display - they missed just two tackles in nearly 100 attempts during the first 70 minutes of action before it was breached late on.
But they were equally adept in attack and their first try, through Tom Brown, owed much to the break of teammate Jack Cuthbert.
With Perpignan hastily closing down the marauding full back, a huge gap opened up for Brown who was left to scamper in from 30 metres.
PLAY OF THE WEEKEND
Victory for Munster over Gloucester never looked in doubt after they eased into a 10-0 lead, with Ian Keatley's clever grubber kick playing Keith Earls for the opening try eight minutes from the break.
Superb approach work with ball in hand from Paul O'Connell and flanker Tommy O'Donnell spread panic in the Gloucester defence.
And it left Keatley with time to find his spot, kicking into space for Earls to finish with determination.
PLAYER OF THE WEEKEND
In what was a far from stellar performance from Leinster, one man to stand out was Jimmy Gopperth who made the most of his chance at fly-half.
The New Zealander dotted down twice and kicked 13 points as Leinster eventually clicked into gear after a slow start that saw them fall 14-0 down.
The former Newcastle Falcons man was at the heart of everything and rightly picked up the man of the match accolade.
