But defeat to fifth-placed Leinster will change the picture completely, with current top two Munster and Glasgow Warriors left to play.
Though Friday's match is not quite winner takes all, Best does not want Ulster's hard work over the past seven months to fall by the wayside.
"We wanted to get to this last phase with our destiny in our own hands and that is where it is," Best told the Belfast Telegraph.
"It is a very tough run-in, but you have to play everyone and beat them if you want to win the competition.
"We've put ourselves in a good position and we'll be confident going into Leinster as it is at home and we're also coming off the back of a good win in Connacht."
