Hines has been named among the replacements for the 3pm kick-off in Edinburgh but his experience and versatility makes it unlikely he will not feature at some stage.
And the Australian-born Scotland star could face up to 10 of his club-mates after Declan Kidney named eight Leinster players in his Ireland XV, with two more on the bench.
If Hines comes on in the second row, he could be pitted against Leinster colleague Leo Cullen - himself a replacement - while if he is required in the back row he may get up close and personal with Jamie Heaslip and Sean O'Brien.
Fellow Leinster regulars Luke Fitzgerald, Brian O'Driscoll, Gordon D'Arcy, Eoin Reddan, Mike Ross and Cian Healy all start for Ireland, while Jonathan Sexton joins Cullen on the replacements bench.
Ospreys wings Nikki Walker and Tommy Bowe also line up on opposing sides, with Walker starting for Scotland and Bowe returning from injury to feature for Ireland.
The pair have been spared the prospect of being in direct combat, however, after both were named on the right wing for their respective teams.
