Moffat's men are currently four points behind the league leaders but victory at Murrayfield could see them overtake the Welsh region and move back into a coveted Play Off spot.
Defeat to Munster last time out saw Edinburgh slip to fifth in the standings and, after a two-week break, Moffat is delighted his side have the chance to regain their place in the top four.
"The Ospreys are top of the league but that can change very, very quickly - we could overtake them at the weekend," said Moffat, who has seen his team win half of their 12 league games so far this season.
"Everyone connected with us knows it's a must-win game.
"Not having played for two weeks since Munster, it's a difficult time of the season.
"We're delighted to have a game, and a game at home. We've not had a home game for a while so it'll be good to play at Murrayfield, hopefully in front of a good crowd and in good conditions.
"We always talk about performance - hopefully if you put in a good performance, you'll win."
Having been beaten 31-10 at the Liberty Stadium back in October, Edinburgh know just how tough this weekend's encounter is likely to be.
Moffat is fully aware of the potential of the former Magners League Champions and he has warned his players they will need to be at their best to record their seventh win of the campaign.
"We know what kind of side the Ospreys are - they like to play a bit of rugby like most teams do," Moffat told Edinburgh's Black and Red Radio.
"They carved us up two or three times down at the Liberty Stadium earlier on this year.
"There's not that many sides that score from first phase but they can do that. We know the danger they have.
"Like most Welsh sides, they're physical, and if you've got players like Jerry Collins and Marty Holah and Filo Tiatia in the back row…we have to be very physical against them. If you don't win that battle then you're struggling all over.
"We know it will be a physical battle - we want to win that and we want to play some good rugby as well."
