The Ospreys and Scarlets both provide four players with Wales skipper Ryan Jones, Andrew Bishop, Duncan Jones and Ian Gough representing the Liberty Stadium outfit.
Scarlets wing Mark Jones is fit-again and the 46-cap star is joined by regional colleagues Dafydd Jones and young guns Daniel Evans and Jonathan Davies.
But captain Jones has warned his side to leave nothing in the tank as Wales aim to finish their two-Test tour of North America on a high at Toyota Park.
The Ospreys No8 has issued a rallying call to his team-mates after demanding one final push against former Ireland coach Eddie O'Sullivan's Eagles before the 2008/09 season draws to a close.
"Everyone is aware we have a five-week break to come but that will start on Sunday morning," said Jones.
"We've got players who are making their first and second appearances for Wales and looking to impress. We need to empty what is left in the tank.
"I was really impressed with the USA against Ireland last weekend and we know they will be big and pretty fierce upfront. We expect nothing less.
"The breakdown will be key as it seems to be in every game now - we can't afford to make any mistakes."
New Gloucester signing Nicky Robinson, who last played for Wales against Argentina in 2006, will form a half-back partnership with Sale Sharks scrum-half Dwayne Peel as two of five changes made by caretaker coach Robin McBryde to face the Eagles, ranked 19th in the world.
Scarlets flier Mark Jones replaces Chris Czekaj on the wing while Gough and Williams are included to bolster the Welsh forward pack with uncapped Sam Warburton named on the bench.
